This presentation addressed the use of PermeaPad® 96-well plates as a high-throughput in vitro tool for assessing drug permeability and predicting oral drug absorption. The principles of apparent permeability (Papp) and the advantages of artificial barriers compared with conventional cellular systems were discussed, with particular emphasis on passive drug transport and formulation screening. Experimental applications of PermeaPad® were presented, including its use for in vitro–in vivo correlation, evaluation of pH-dependent permeability, and simulation of gastrointestinal conditions. The presentation highlighted the potential of high-throughput permeability testing to accelerate pharmaceutical development, improve mechanistic understanding of drug absorption, provide parameters for in silico modelling, and reduce reliance on animal experiments in accordance with the 3Rs principle.
