Generation of superoxide anion (O2−) by phagocytizing bovine blood granulocytes and extracellular release of the anion were simultaneously assayed by measuring oxygen consumption and O2−-dependent ferricytochrome c reduction. O2− release, but not generation, was markedly inhibited by 4-acetamido-4′-isothiocyano-stilbene-2,2′-disulfonate and by 1-anilino-8-naphtalene sulfonate, with an I50 of about 50 μM. Inhibition of O2− release was also observed when a portion of NaCl in the cell suspending medium was replaced by NaNO3 or NaSCN or Na salicylate, with the anions acting in the following order of potency: salicylate > SCN− > NO3−.
