Theidentificationofdifferentpoolsofcardiacprogenitorcellsresidentintheadultmammalianheartopenedanewerainheartregenerationasameanstorestorethelossoffunctionalcardiactissueandovercomethelimitedavailabilityofdonororgans.Indeed,residentstemcellsarebelievedtoparticipatetotissuehomeostasisandrenewalinhealthyanddamagedmyocardiumalthoughtheiractualcontributiontotheseprocessesremainunclear.ThepooroutcomeintermsofcardiacregenerationfollowingtissuedamagepointoutattheneedforadeeperunderstandingofthemolecularmechanismscontrollingCPCbehaviorandfatedeterminationbeforenewtherapeuticstrategiescanbedeveloped.Theregulationofcardiacresidentstemcellfateandfunctionislikelytoresultfromtheinterplaybetweenpleiotropicsignalingpathwaysaswellastissue-andcell-specificregulators.Suchamodularinteraction—whichhasalreadybeendescribedinthenucleusofanumberofdifferentcellswheretranscriptionalcomplexesformtoactivatespecificgeneprograms—wouldaccountfortheuniqueresponsesofcardiacprogenitorstogeneralandtissue-specificstimuli.Thestudyofthemoleculardeterminantsinvolvedincardiacstem/progenitorcellregulatorymechanismsmayshedlightontheprocessesofcardiachomeostasisinhealthanddiseaseandthusprovidecluesontheactualfeasibilityofcardiaccelltherapythroughtissue-specificprogenitors.
